铁死亡(Ferroptosis)是一种铁依赖形式的非凋亡性细胞死亡。Ferrostatin-1(Fer-1)是一种有效的、选择性的铁死亡抑制剂。HT-1080细胞中,Fer-1(0.5μM)抑制铁死亡激活剂Erastin(Cat#:NBS5879,EC50=60nM)和RSL3诱导的细胞死亡,但对过氧化氢、鱼藤酮,或星孢菌素诱导的细胞死亡没有效果。Fer-1抑制Erastin诱导的活性氧生成和脂质过氧化。在基于细胞水平的亨廷顿病、脑室周围白质软化症(PVL)和近端肾小管损伤模型中,Fer-1以剂量依赖性的方式提高细胞存活率。大鼠海马器官型脑片培养模型中,Fer-1(2μM)抑制L-谷氨酸盐诱导的细胞死亡。Fer-1还能通过捕获过氧自由基的方式阻抑脂质自氧化。

实验方法(Assay):HT-22 cells were seeded in 6-well culture plate (2 × 105 cells/well). Glutamate (1.25, 2.5, 5, 10, 20 mM) was applied to HT-22 cells for 12, 24, 36, and 48 hours. HT-22 cells were cultured with different concentrations of ferrostatin-1 (3, 6 and 12μM) for 16 hours prior to exposure to 5 mM glutamate. After 24 hours, the subsequent analyses were conducted.
结果说明(Result):Ferrostatin-1 dose-dependently reduced cell death, with a maximum effect at a dose of 12 μM, but had no significant effect on the survival of HT-22 cells deprived of glutamate.

体内研究(动物模型):
动物模型(Animal Model):Male C57BL/6 mice
实验方法(Assay):The LPS-induced ALI model was induced by instilling intratracheally 50 μl of LPS solution (0.2 g/L), then Fer-1 (0.8 mg/kg) was administered after LPS challenge via tail vein injection. The Fer-1 was dissolved in DMSO first, and diluted with 0.9% NaCl. The final concentration of Fer-1 and DMSO was 0.2 mg/ml and 0.1% respectively. After the treatments for 16 h, the mice in each group were euthanized and bronchoalveolar lavage (BAL) fluid was collected via lung lavage.
