(1S,3R)-RSL3是一种谷胱甘肽过氧化物酶4(GPX4) 抑制剂,诱导致命性的脂质氢过氧化物累积和铁死亡。与野生型BJeHLT细胞(EC50 = 2 µM)相比,选择性作用于表达HRASV12的BJeHLT细胞(EC50 = 0.01 µM)。各种小鼠移植瘤模型中,(1S,3R)-RSL3减少肿瘤体积,其中带弥漫大B细胞淋巴瘤和肾细胞癌的模型最敏感。

体外研究(细胞水平):
细胞类型(Cell Type):A549 and H2122 cells
实验方法(Assay):A549 cells were treated withdimethyl sulfoxide (DMSO) or RSL3 (4 μM) for 12 hand H2122 cells were treated withDMSO or RSL3 (8 μM) for 12 h and then subjected to CCK-8 assay.
体内研究(动物模型):
动物模型(Animal Model):10-week-old male nude micewere subcutaneously injected with A549 cells (100 μL containing 5 × 106 cells/injection) and monitored for tumor cell xenografts to reach approximately 100 mm3.
实验方法(Assay):The mice were then divided into two groups (n = 5), the RSL3 treatment (100 mg/kg; dissolved in 5% dimethyl sulfoxide/corn oil; administrated intratumorally twice a day for one week) and control (5% dimethyl sulfoxide/corn oil only) groups. After that, the mice were sacrificed, and tumor weight was measured respectively.
